《刺客信条：兄弟会》将在2010年圣诞商战期间发售，平台为Xbox360，PS3和PC。游戏是由育碧蒙特利尔工作室开发。在单人模式中，玩家将再度化身为传奇杀手的前作主角Ezio／エンツィオ，并为了与圣殿骑士团战斗而前往罗马。另外较于前作都是在描述单独行动的内容，在这次的系列最新作当中，则将以主角率领伙伴一起行动为主，玩家们在游戏里可以对各伙伴下达命令，并通过他们的协助来完成任务。在多人模式方面，会有多种职业可选择，而且各职业都具有专属武器和刺杀技能。
紧接前作《刺客信条II》 ，酒保戴斯蒙德·迈尔斯（Desmond Miles）与其余三名现代刺客露西·斯提尔曼（Lucy Stillman）, 肖恩·黑斯廷斯（Shaun Hastings）瑞贝卡·科瑞尔（Rebecca Crane）逃离圣殿骑士团的追击后到达意大利城市蒙特里久尼。以已荒废的奥迪托雷庄园地下室作为新的藏身之所，Desmond再次使用基因记忆追溯机器“Animus 2.0”，再度进入其刺客祖先艾齐奥·奥迪托雷·德·弗洛伦萨 （艾齐奥 Auditore da Firenze）的记忆。
